چکیده

PCR-based analysis of skeletonized human remains is a common aspect in both forensic identification as well Ancient DNA research. In this, areas not merely utilize very similar methodology, but also share the same problems regarding quantity and quality recovered presence inhibitory substances samples from excavated remains. To enable amplification based remains, development optimized extraction procedures thus critical factor areas. The study here presents an protocol for ancient using Chelex-100, which proved to be effective yielding amplifiable extracts sample material after centuries soil environment, consequently have high inhibitor content overall limited preservation. Success optimization strategies utilized shown significantly improved outcomes compared predecessor method

Authenticating DNA Extracted From Ancient Skeletal Remains

The survival of DNA, the most informative biological molecule, for periods of at least several thousand years in bone was demonstrated more than four years ago. However, difficulties with authenticating ancient DNA have made diagenetic studies problematic. It is therefore essential that these problems be overcome before the question of DNA survival can be addressed. Here we describe our work wi...

Comparison of two ancient DNA extraction protocols for skeletal remains from tropical environments.

OBJECTIVES The tropics harbor a large part of the world's biodiversity and have a long history of human habitation. However, paleogenomics research in these climates has been constrained so far by poor ancient DNA yields. Here we compare the performance of two DNA extraction methods on ancient samples of teeth and petrous portions excavated from tropical and semi-tropical sites in Tanzania, Mex...

DNA preservation: a microsatellite-DNA study on ancient skeletal remains.

To determine the effect of environmental factors on the preservation of DNA, archeological teeth of approximately similar age but greatly differing site milieu were examined for DNA content. The complex relational system of locational milieu of the samples was reduced to its essential and, at the same time, easily measurable factors. These are temperature, humidity, pH value, the geochemical pr...
